 DMEM 高不含酚紅谷氨酰胺丙酮酸鈉 細(xì)胞培養(yǎng)

DMEM是 dulbecco's modified eagle medium的縮寫,是一種含各種氨基酸和葡萄糖的培養(yǎng)基，是在MEM培養(yǎng)基的基礎(chǔ)上研制的。與MEM比較增加了各種成分用量，同時(shí)又分為高糖型（低于4500mg/L）和低糖型(低于1000mg/L)。高糖型有利于細(xì)胞停泊于一個(gè)位置生長(zhǎng)，適于生長(zhǎng)較快、附著較困難腫瘤細(xì)胞等。所以其特點(diǎn)主要包括以下：
（1）氨基酸含量為依格爾培養(yǎng)基的2倍,且含有非必需氨基酸,如甘氨酸等；
（2）維生素含量為依格爾培養(yǎng)基的4倍；
（3）含有糖酵解途徑中的重要物質(zhì)——丙酮酸；
（4）含有微量的鐵離子.Eagle來自該培養(yǎng)基的發(fā)明人Harry Eagle

1640和DMEM培養(yǎng)基的區(qū)別

1640培養(yǎng)基和DMEM培養(yǎng)基在組分和用途上有區(qū)別，主要是組分上的區(qū)別，查各自的配方表可知。用途上這兩種培養(yǎng)基應(yīng)用都很廣泛，且都適合很多種細(xì)胞的培養(yǎng)。
    RPMI 1640主要用于懸浮細(xì)胞培養(yǎng)。可適用于哺乳動(dòng)物、特殊造血細(xì)胞、正常或惡性增生的白細(xì)胞，雜交瘤細(xì)胞的培養(yǎng)。其它像K-562、HL-60、IM-9等成淋巴細(xì)胞、T細(xì)胞淋巴瘤細(xì)胞以及HCT-15上皮細(xì)胞等均可參考使用。
    DMEM適用于貼壁細(xì)胞生長(zhǎng)，如各種實(shí)體瘤貼壁細(xì)胞。
